I got it working finally, and want to let you know what i did:
I made an Compressed Image File of the USB-Key (img.gz), done by the tool "USB Image Tool". Just place this file where you want, such as in _ISO/UTILITIES.
Easy2Boot is doing all by itself.

" .. E2B\grub4dos automatically detects if a file has been compressed to gzip or LZMA format. The file extension is not used by E2B to determine if a file is compressed or not. For instance, a LZMA-compressed DOS floppy image can be named .ima or .imagz."
